Effect of Thidiazuron (tdz) on in Vitro Micropropagation of Solanum Tuberosum L. Cvs. Desiree and Cardinal

چکیده
Thidiazuron (1-phenyl-3(1,2,3Thiadiazol-5-yl) urea; TDZ) is one of the several substituted ureas that have been investigated recently for their cytokinin-like activity. TDZ is known to be more active than zeatin for stimulating the growth when added to a tissue culture medium at a low concentration. In this study, effect of TDZ on several In vitro growth parameters in Solanum tuberosum cvs. Desiree and Cardinal were observed. Shoot apices (1.0cm each) from both the cultivars were separately inoculated on full strength MS basal media as well as on MS full strength supplemented with different concentrations of TDZ (10, 10 or 10 M) thus forming four combinations. Results were recorded for shoot length, shoot number, root length, root number, number of nodes, fresh and dry weight of plants after 30 days of explant inoculation. MS full strength medium was found to be the best for In vitro micropropagation. However, TDZ did have an influence on the studied growth parameters, which have been discussed and interpreted herein.

In Vitro Micropropagation of the aquarium plant Ludwigia repens
Apical meristems, first, second and third-fourth axillary buds of Ludwigia repens were cultured on Murashige and Skoog (MS) medium containing various concentrations of 6-benzylaminopurine (BAP), thidiazuron (TDZ) and αnaphthaleneacetic acid (NAA) for micropropagation. Micropropagation was best achieved from apical meristem on MS medium containing 0.05 mg dm TDZ and 0.1 mg dm NAA. It was noted t...

Genetic variation due to somaclonal variation in micropropagated plants is a beneficial phenomenon for crop improvement. Genetic integrity of the plants derived through micropropagation becomes crucial if genetic transformation studies have to be carried out. Somaclonal variation in tissue culture is a common phenomenon which makes it mandatory to check for genetic stability of plants. Hypocoty...

In vitro budinduction and shoot regeneration of Lantana camara L. was investigated using axillary nodal explants. Multiple shoot buds were induced on Murashige and Skoog (MS) media supplemented with 0, 2, 4 and 8 mg/l of 6-benzylaminopurine (BAP) or thidiazuron (TDZ) for proliferation and 0, 0.25 and 0.5 mg/l indole-3 butyric acid (IBA) for rooting. After 4 weeks, a maximum number of shoot buds...
متن کاملPlant Regeneration from in Vitro-selected Salt Tolerant Callus Cultures of Solanum Tuberosum L
This research work reports In vitro direct selection of salt-tolerant callus cultures and subsequent plant regeneration in two potato cultivars (Cardinal and Desiree). Results have shown more than 50% reduction in relative fresh callus mass in the two potato cultivars exposed to 120 mM NaCl. Callus morphology correspondingly changed from off-white to blackishbrown at 120 mM to acutely-necrotic ...
متن کاملThe Effects of Different Concentrations of Nitrogen Sources on Growth of Micropropagated Potato Cultivars
The use of nitrogen in nutrient media is essential for growth and helps identify highly productive media for potato (Solanum tuberosum L.) micropropagation and adventitious shoot formation. Three potato cultivars (Agria, Marfona and Savalan cultivars) were examined for their growth response with medium containing four different levels of KNO3 (1, 1.25, 1.5 and 1.75 times as much 1.9 g l-1) and ...
